Protocol Development Service — Custom TC Protocol for Your Target Species

The Phytolabs Protocol Development Service provides customised, experimentally validated tissue culture protocols for plant species or plant products for which no established protocol exists, or where existing published protocols have failed to produce acceptable results in your specific genotypes or production conditions. Our plant biotechnology team designs, executes, and optimises complete TC protocols covering explant selection and preparation, surface sterilisation, medium composition, growth regulator regime, incubation conditions, subculture intervals, and acclimatisation procedures, delivering a documented, reproducible protocol validated in your target species.

Protocol development is needed when working with novel species not previously cultured (endemic species, new crop species, specialised medicinal plants), recalcitrant varieties of established crop species (specific banana cultivars with poor Stage II multiplication rates, rose cultivars resistant to standard BAP protocols), and when existing published protocols fail to reproduce in the client’s specific explant source material or laboratory conditions. The service is also valuable for organisations requiring protocols for regulatory submissions (variety registration, plant breeders’ rights) or for protocols needing specific documentation standards for ISO-certified commercial operations.

The development process follows a systematic experimental approach: initial literature review of closest-related species protocols; explant source evaluation and seasonal timing optimisation; surface sterilisation screening across three or more sterilant combinations and timings; base medium screening (MS, B5, WPM, other appropriate formulations); growth regulator factorial screening for each culture stage; physical condition optimisation (photoperiod, temperature, humidity, vessel type); and scale-up validation to confirm that protocols developed at research scale (6-well plates, Magenta vessels) translate reliably to commercial containers (glass jars, temporary immersion systems). Full statistical analysis of experimental data is provided.

Deliverables include: a complete written protocol with all media compositions, growth regulator concentrations, timing, and procedural steps; supporting experimental data and statistical analysis; recommended equipment and consumables list; and a one-day on-site training session to transfer the protocol to the client’s team. The protocol includes explant preparation, Stage I establishment, Stage II multiplication, Stage III rooting, and Stage IV hardening stages as applicable to the species.
