⚗️ KOH vs NaOH in TC
Most TC protocols are indifferent to whether you use KOH or NaOH for pH adjustment — the amount added is tiny relative to total medium composition. However, KOH is recommended when: (1) the medium already has high Na⁺ (e.g., high-ammonium formulations); (2) studying sodium sensitivity or sodium exclusion in halophyte/glycophyte comparisons; (3) the published protocol specifically uses KOH (follow the protocol). For routine MS pH adjustment either is fine.
💡 Prepare 1M KOH: dissolve 5.6 g KOH pellets in 100 mL distilled water. Same hygroscopic caution as NaOH — keep tightly sealed.
