๐ก Why proline in TC?
Proline serves three roles in TC: (1) osmotic adjustment โ proline accumulation is a universal stress response; exogenous proline improves culture recovery under osmotic stress; (2) pollen wall synthesis โ sporopollenin and pollen wall proteins are proline-rich; (3) somatic embryogenesis โ proline induces embryogenic competence in some cereals (wheat, barley) and legumes by activating stress-related SE pathways.
๐พ Working concentrations
| Use | Concentration | Notes |
|---|---|---|
| Cereal anther culture (SE induction) | 100โ500 mg/L | Wheat, barley protocols |
| Pollen germination medium | 100โ300 mg/L | Improves germination % |
| Somatic embryogenesis (legumes) | 100โ500 mg/L | Soybean, alfalfa |
| Osmotic stress tolerance experiments | 50โ500 mg/L | Dose-response |
