Description

๐Ÿ–ค Activated Charcoal (TC Grade) โ€” high-surface-area adsorption agent (500โ€“1500 mยฒ/g); removes inhibitory compounds from medium and spent metabolites secreted by cultures. Essential for orchid PLB development, rooting of some woody species, and somatic embryo germination.

๐Ÿงช CAS 7440-44-0 ๐Ÿ“ฆ Form Black powder
๐Ÿ”ฌ Grade TC / Adsorption ๐ŸŒก๏ธ Storage RT, sealed, dry

๐Ÿ–ค What activated charcoal does in TC

Charcoal adsorbs: phenolics (browning compounds), ethylene (gaseous inhibitor that accumulates in sealed containers), 5-hydroxymethylfurfural (autoclaving artefact), spent hormone metabolites, and possibly low levels of exogenous inhibitors in crude agar. This “clean medium effect” dramatically improves: orchid PLB development, somatic embryo germination, and rooting of recalcitrant woody species.

๐Ÿ–ค Working concentrations

Use Concentration Notes
Orchid PLB development & maturation 0.5โ€“2 g/L Removes ethylene; promotes differentiation
Rooting of recalcitrant woody species 1โ€“3 g/L WPM + IBA + charcoal is classic
SE germination / conversion medium 1โ€“2 g/L Removes inhibitory compounds
Callus with browning problem 0.5โ€“1 g/L Adsorbs phenolic exudates
Banana Stage III (rooting) 1โ€“2 g/L Widely used in commercial production

โš ๏ธ Activated charcoal also adsorbs plant growth regulators โ€” especially IAA, GAโ‚ƒ, and zeatin. Do not use charcoal-containing medium when precise hormone concentrations are needed. BAP and IBA are less affected, but test your specific hormone.
