Description

🌿 Naphthaleneacetic Acid (NAA) — the most stable synthetic auxin for TC. More persistent than IAA (not oxidised by IAA oxidase), more reliable than 2,4-D for rooting.

🧪 CAS 86-87-3 ⚖️ MW 186.21 g/mol
🔬 Grade TC / Research 🌡️ Storage RT, dark, dry; stock at 4 °C

⚗️ How it works

NAA mimics IAA at TIR1/AFB auxin receptors, promoting cell elongation, division, and root initiation. Because it is not degraded by IAA oxidase, its effect is more prolonged and predictable than natural auxin — a major advantage in a controlled TC medium.

🌱 Working concentrations

Use Concentration Notes
Root induction on shoots 0.5–3 mg/L ½ MS, no cytokinin
Callus induction (with BAP) 0.1–2 mg/L Higher than BAP → more callus
Shoot culture maintenance 0.01–0.1 mg/L Low conc reduces callus base
Adventitious root induction 1–5 mg/L Pulse treatment effective
Embryogenic callus (some spp) 2–5 mg/L With low cytokinin
Banana Stage II supplement 0.1–0.3 mg/L With high BAP

🧪 Stock solution prep

Dissolve in a few drops of 1M NaOH or 70% ethanol, dilute to volume. Prepare 1 mg/mL stock, filter-sterilise, store at 4 °C.

💡 NAA at >1 mg/L without cytokinin will drive rooting in most shoot cultures. If roots appear during Stage II multiplication, reduce NAA or remove it entirely.
