Description
๐งซ Gelrite (Deacylated Gellan Gum) โ a microbially-produced polysaccharide gelling agent that gives a crystal-clear gel; allows visual contamination detection and is the preferred gelling agent for somatic embryogenesis and conifer TC.
| ๐งช CAS | 71010-52-1 | ๐ฌ Grade | TC / Tissue Culture |
| โ๏ธ Gel conc | 1.5โ3 g/L (vs 6โ8 g/L for agar) |
๐ก Advantages over agar
- โ Crystal-clear gel โ contamination visible immediately
- โ Lower use concentration (2โ3 g/L vs 7 g/L for agar) โ lower salt/inhibitor carryover
- โ More defined composition (single polysaccharide vs agar complexity)
- โ Better embryo development in SE protocols โ softer texture
- โ ๏ธ Higher cost per gram than agar
- โ ๏ธ Requires divalent cations (Mgยฒโบ, Caยฒโบ) for gel formation โ present in most TC media
๐งซ Working concentrations
| Use | Concentration | Notes |
|---|---|---|
| Somatic embryogenesis (standard) | 2โ3 g/L | Conifer, carrot, banana SE |
| Shoot culture (alternative to agar) | 2โ2.5 g/L | Clearer medium, better contamination QC |
| Callus induction | 2โ2.5 g/L | |
| Root induction | 1.5โ2 g/L | Softer gel for easier root penetration |
โ ๏ธ Gelrite requires divalent cations to gel. In media with very low Caยฒโบ/Mgยฒโบ (modified formulations), gel strength may be inadequate โ add 1 mM CaClโ if gel is too soft.
