Sucrose is the preferred TC carbon source because it is the natural phloem transport sugar β€” plants are adapted to import it. Glucose is used when: (1) studying hexose vs sucrose signalling (different sugar sensing pathways), (2) protoplast and single-cell cultures that have low invertase activity, (3) anther culture (some protocols: pollen absorbs glucose directly), or (4) when sucrose hydrolysis during autoclaving must be avoided.
