Description
🍋 Ascorbic Acid (Vitamin C) — antioxidant for TC. Scavenges reactive oxygen species and prevents quinone-mediated browning in polyphenol-rich explants. Works synergistically with PVP-40.
| 🧪 CAS | 50-81-7 | ⚖️ MW | 176.12 g/mol |
| 🔬 Grade | TC / Analytical | 🌡️ Storage | 4 °C, dark, sealed; degrades on exposure to air |
🛡️ Anti-browning applications
| Use | Concentration | Notes |
|---|---|---|
| Explant wash / soak solution | 100–200 mg/L in distilled water | Pre-culture soak 30–60 min |
| Anti-browning supplement in medium | 50–100 mg/L | Combine with PVP for recalcitrant spp |
| Dissection buffer (for sterile dissection) | 0.1–0.2% ascorbic acid | Keep explants moist & protected |
| Feeder layer medium supplement | 50 mg/L | Reduces oxidative stress |
💡 Ascorbic acid in medium must be filter-sterilised — it degrades significantly during autoclaving. Prepare a 10 mg/mL stock (1%), filter through 0.22 µm, and add to cooled autoclaved medium.
⚠️ Ascorbic acid in solution oxidises to dehydroascorbic acid within hours at RT. Prepare fresh, use immediately, or keep stock at 4 °C in the dark for max 1 week.
